TERT mRNA is the messenger RNA encoding the enzyme telomerase. The proven science behind the TERT mRNA is supported by the founding research of Rejuvenation Technologies. 

Telomere shortening is a natural process that occurs at an accelerated rate in research cell lines. Cell lines eventually reach a proliferative cap when their telomeres become too short and enter cellular senescence. This leads to poorly or non-reproductive cell lines and subsequently repurchasing of the line. Transient delivery of Rejuvenation’s TERT mRNA leads to an unbiased extension of the telomere ends within a cell.

By repairing and extending telomere ends, cells continue to proliferate as usual. This means that telomeres continue to shorten after repair, thus leading to eventual senescence. There is no known limit of treatment per cell line.

About – TERT mRNA, Telomere Shortening and its Function

TERT mRNA, messenger RNA which encodes the enzyme telomerase reverse transcriptase (TERT, was developed at Stanford University[superscript 1 that links to reference] as a tool to safely improve the quality and proliferation capacity of cells for cell culture, cell therapies, tissue engineering, and in vivo therapeutics. Rejuvenation’s TERT mRNA is synthetic mRNA synthesized in vitro using naturally occurring modified nucleosides that make the mRNA hypoimmunogenic when delivered to cells using standard transfection methods.

TERT mRNA is naturally produced in transient bursts by cells in the body and enables cells to delay replicative senescence caused by short telomeres. Telomere shortening is a natural process that occurs when cells divide. Telomeres shorten at an accelerated rate when cells are constantly dividing as in culture. When their telomeres become too short, cells reach a proliferative limit and enter replicative senescence or die. This leads to decreased quality and utility of cells, and in the case of primary cells necessitates re-isolating or repurchasing. 

Unlike other industry-standard treatments, TERT mRNA does not immortalize cells. A single dose of TERT mRNA to cells, administered in about 15 minutes, typically extends telomeres by several hundred base pairs (bp). This is often sufficient to increase the proliferative capacity of cells by up to 15 population doublings (PD). During treatment, the mRNA is transcribed into a telomerase protein that goes to the nucleus and extends the telomeres. The TERT mRNA and protein are then degraded within hours and days, respectively. Immediately after the treatment ends, the telomeres resume shortening at their normal rate.

How much TERT mRNA do I need? 

We recommend that you use 1 ug of TERT mRNA per 1ml of medium. The amount or type of cells within the medium is irrelevant.

What cell lines does TERT mRNA work with? 

We have not yet identified a cell line for research purposes only that does not respond to treatment. The amount of proliferation extension may differ from line to line, ranging from 15-25 additional population doublings. 

What should I use to deliver the TERT mRNA?

We highly recommend utilizing Thermo Scientific’s MessengerMax for delivery to cells in most applications (sold separately). Please follow the instructions provided by Thermo Scientific while formulating.

How do I know if my cell line’s proliferation capacity has increased? 

The cheapest way to confirm success is to proliferate your cell line past its usual limit. However, you can analyze your cell’s telomere length before and after to confirm the product’s effects immediately.

Our product must be stored and transported at -80C. The product will be shipped in an insulated shipping box via FedEx Overnight shipping. Upon delivery, we ask that you please store the product within a -80C freezer immediately to preserve the mRNA’s integrity.

Mode of action

When transfected into cells, TERT mRNA is translated into telomerase protein that is translocated to the nucleus and rapidly extends telomeres in most cell types. The mRNA is degraded within hours and the protein is degraded within days. During this brief period, the telomeres are extended rapidly, typically by hundreds of base pairs (bp). This is equivalent to reversing years of telomere shortening during normal ageing.
